This is a rare moment of genuine vulnerability for Sheldon, and it subtly sets up his future character (the older Sheldon in TBBT still fears failure but hides it behind arrogance).

The episode opens with Sheldon and Dr. John Sturgis in Sheldon’s dorm room at East Texas Tech. Sheldon is struggling with a particularly difficult problem in — something beyond even his usual brilliance. Sturgis tries to help, but Sheldon is frustrated, saying, “This nut is tougher than I anticipated.” Sturgis reminds him that even Einstein had problems that took years to solve.
